At the first interview, I met with the hiring manager and the woman whose position I was going to backfill. Relaxed but professional.
Second interview I met with SVP and counterpart. Also somewhat relaxed, but more formal than the previous interview. Asked - what would you like me to take away from this interview? Why are you interested in this position over the other positions you're received offers from?
